 ng mechanisms regulating the release of\nIL-1b and the importance of IL-1b
  in numerous inflammatory\ndiseases. Human genome analysis led to the disc
 overy of a novel\nfamily of NLR proteins involved in innate immunity\, sev
 eral of\nwhich form intracellular complexes known as inflammasomes\, which
 \nrespond to pathogens\, non pathogenic inflammatory stimuli\, and\nintern
 al danger signals. Advances in human genetics have revealed a\nprincipal r
 ole for IL-1b in the pathogenesis of recurrent fever\ndisorders and furthe
 r studies have expanded the autoinflammatory\ndisease family beyond rare M
 endelian syndromes to include common\nconditions such as gout and occupati
 onal lung disease. With the\nadvent of IL-1-targeted therapy\, diseases as
 sociated with excess\nIL-1b can now be treated effectively.\n
